Solid Phase Extraction SPE

Solid phase extraction (SPE)-

It is a technique designed for rapid, selective sample preparation, purification prior to chromatography analysis.
SPE provides the sample cleanup, recovery, and concentration necessary for accurate quantitative analysis using liquid chromatography principles to control selectivity.
The multitude of available phase chemistry can be packed into an array. This array are in the form of hardware formats such as glass tubes, 48, 96-well, 144, plates, and Cartridge tubes (dispersive SPE). This  can be processed using dedicated vacuum manifold accessories.

(SPE) is a technique by which compounds that are dissolved in a liquid mixture are separated from other compounds in the mixture. Some Analytical laboratories use solid phase extraction SPE  to concentrate and purify the samples for analysis. Though including urine, blood, water, beverages, soil, and animal tissue.

Many of the adsorbents/materials are the same as in chromatography methods. But Solid Phase Extraction SPE is distinctive, with the aims separate from chromatography. So it has a unique niche in modern chemical science.

SPE and Chromatography :

SPE is a method of chromatography, except in the broadest, simplest sense. It is an extractive technique, taking advantage of large differences in the Keq, or equilibrium constant. It is of mixture components between the solid phase and the mobile phase resulting. This is for a well-designed and executed separation, in a bulk separation of one or more of the mixture components.

24-port SPE (Solid Phase Extraction) vacuum manifold is a laboratory device designed for the simultaneous processing of up to 24 samples during solid phase extraction. It utilizes vacuum to draw solvents and samples through SPE cartridges, enabling efficient sample preparation for various analytical techniques. Key specifications include the number of ports (24), the type of valves for flow control (often screw-type or push-type), the material of construction (often glass or polycarbonate), and the vacuum range (e.g., 0-20 inches Hg or 0-0.08 MPa). Key Features and Specifications:
  • 24 Ports:Allows for the parallel processing of 24 samples, significantly increasing throughput compared to single-sample methods.
  • Flow Control:Individual valves (screw-type or push-type) are used to precisely control the flow rate through each SPE cartridge, preventing drying out and ensuring uniform processing.
  • Vacuum System:A vacuum source (e.g., vacuum pump) is connected to the manifold, creating a pressure differential that draws solvents and samples through the cartridges.
  • Material:Often constructed from durable materials like borosilicate glass or polycarbonate, with polypropylene or PTFE fittings, to withstand solvents and chemicals.
  • Vacuum Range:Typically operates within a specific vacuum range, such as 0-20 inches of mercury (Hg) or 0-0.08 MPa.
  • Collection Rack:A removable rack inside the manifold holds collection tubes or vials to collect the processed samples.
  • Pressure Gauge:Many manifolds include a pressure gauge to monitor the vacuum level during operation.
  • Safety Features:Some manifolds may include features like solvent-resistant vacuum bleed valves and CE certification for safety.
  • Optional Accessories:Options like long-stem flow control valves, disposable liners, and different collection racks may be available for specific applications.
Applications:
  • Environmental Analysis:Used to extract and concentrate pollutants, pesticides, and other contaminants from water, soil, and air samples.
  • Pharmaceutical Analysis:Applied in pharmaceutical research and quality control for isolating drug compounds, metabolites, and impurities from biological fluids and other samples.
  • Food and Beverage Analysis:Used for analyzing contaminants, additives, and other compounds in food and beverage samples.
  • Clinical Chemistry:Applicable for sample preparation in clinical diagnostics and biomarker analysis.

Solid Phase Extraction (SPE) vacuum manifold is a laboratory apparatus used to prepare samples for analysis by isolating and purifying specific compounds. It uses vacuum pressure to draw liquid samples through SPE cartridges, retaining desired analytes while removing unwanted substances. This process enhances the accuracy and sensitivity of subsequent analytical techniques like chromatography and mass spectrometry. Key Applications:

  • Environmental Analysis: Monitoring pollutants in water, soil, and air.
  • Food Safety: Detecting pesticide residues and contaminants in food products.
  • Clinical Diagnostics: Preparing biological samples for analysis of drugs, hormones, and other biomarkers.
  • Pharmaceutical Analysis: Purifying drug compounds from complex matrices in drug discovery and development.
  • Chemical Analysis: Isolating and purifying specific chemicals from various mixtures for further study.

How it Works:

  1. 1. Sorbent Activation:The SPE cartridge is prepared by passing a conditioning solution through it to activate the sorbent material.
  2. 2. Sample Loading:The liquid sample is passed through the cartridge, and the target compounds bind to the sorbent.
  3. 3. Washing:Interfering compounds are washed away using appropriate solvents.
  4. 4. Elution:The target compounds are then eluted from the sorbent using a stronger solvent, collecting the purified sample.
  5. 5. Vacuum Application:The vacuum manifold provides the force to pull the liquid through the cartridge, controlling the flow rate and ensuring efficient extraction.

Benefits:

  • High Sample Throughput:Allows for processing multiple samples simultaneously, increasing efficiency.
  • Reduced Solvent Consumption:Optimized extraction procedures minimize solvent usage compared to traditional liquid-liquid extraction.
  • Improved Sensitivity:Removes interfering substances, enhancing the detection of target analytes in subsequent analyses.
